M521 is a Y chromosome mutation used as a UEP to define a branch of E-M78, with the phylogenetic clade name of E1b1b1a5.

It's discovery was announced in Battaglia et al. (2008) after a study of 1206 people from 17 population samples, mainly from Southeastern Europe.

It was found in two Greek individuals out of 92 tested, and appears to have been absent in two other individuals in that study who also initially appeared to be E-M78* in that study: 1 out of 55 Albanians, and 1 out of 64 Albanian speakers from the Former Yugoslavian Republic of Macedonia (FYROM).

For more detailed information, see the article about the clade E-M521.

The authors described the mutation as "AC012068.5: g.5219 C->T". They say it was discovered while typing V12.
